There's a shuttle service to and from the car park. The paths to and around the gardens are pretty good, but the uphill bits would be hard work with a wheelchair. Tatton Park would be easier and I know that when my sister had a broken leg, we pushed her round Dunham Massey in a wheelchair - now there's a really special garden - wonderful rhododendrons and azaleas in late spring, early summer, and, in the summer, an amazing variety of hydrangeas.
